DSIP Mechanism: What Is and Is Not Known

The mechanism of DSIP is unresolved. No gene, precursor protein or receptor for DSIP has been isolated, and its natural biological role remains obscure.

What is established?

DSIP has a defined chemical structure, and synthetic DSIP produced EEG changes in a 1977 rabbit study.[1], [9]

DSIP-like immunoreactivity has been detected in the hypothalamus, pituitary, gut, adrenal medulla and pancreas of various species, though immunohistochemistry does not prove natural release.

What hypotheses have been proposed?

Hypotheses include a role as a neuroendocrine modulator and the existence of a related DSIP-like peptide responsible for the observed activity.[9]

Kovalzon and Strekalova hypothesized a DSIP-like peptide, partly because DSIP-like immunoreactivity appears in tissues not particularly relevant to sleep regulation and because some analogues outperformed DSIP itself for slow-wave sleep in rabbits and rats.

Why does stability matter?

DSIP shows low molecular stability and degrades rapidly after administration from outside the body.[9]

This complicates interpretation of administration studies, because effects may depend on degradation products or analogues rather than intact DSIP.
