What is Semaglutide?

Semaglutide is a lab-modified version of a hormone your gut naturally releases after eating, called GLP-1. Normally, GLP-1 breaks down in minutes. Scientists modified it so it lasts about a week in your body. Think of it like upgrading a candle that burns for 5 minutes into one that burns for 7 days — same type of light, just engineered to last much longer. It tells your brain you are full and helps regulate blood sugar.

Mechanism of Action

Semaglutide activates the GLP-1 receptor, mimicking the incretin effect: stimulating insulin secretion, suppressing glucagon, slowing gastric emptying, and acting on hypothalamic appetite centers to reduce food intake. Its albumin-binding fatty acid chain extends its half-life to approximately 7 days.

Research Evidence

Human Evidence

Human Evidence

Extensive human clinical trial data across multiple Phase III programs (SUSTAIN, PIONEER, STEP trials). FDA-approved as Ozempic® (injectable, diabetes), Rybelsus® (oral, diabetes), and Wegovy® (injectable, obesity). Thousands of patients studied across dozens of clinical trials.

Animal Evidence

Animal Evidence

Preclinical animal pharmacology established the mechanism and safety profile before human trials.

Laboratory Evidence

In-Vitro / Lab

Comprehensive receptor binding studies, pharmacokinetic modeling, and structure-activity relationship research.

Frequently Asked Questions

What is the difference between Ozempic and Wegovy?

Both contain semaglutide. Ozempic is approved for type 2 diabetes (doses up to 2mg/week). Wegovy is approved for chronic weight management (dose of 2.4mg/week). Same molecule, different approved indications and dosing.

Is semaglutide a peptide?

Yes. Semaglutide is a modified peptide — a 31-amino-acid analog of human GLP-1 with specific amino acid substitutions and a fatty acid chain that extends its half-life from minutes to approximately one week.

Delivery Methods Studied

  • Injectable (subcutaneous, once weekly)
  • Oral tablet (Rybelsus®)

Stability Notes

Highly stable due to engineered modifications (fatty acid chain, amino acid substitutions). Commercial products stable at room temperature for defined periods. Oral formulation uses SNAC absorption enhancer.
