Developed from ACTH(4-7) Fragment

Semax was developed at Moscow State University by taking the ACTH(4-7) fragment (which retains neurotrophic activity without cortisol effects) and adding a Pro-Gly-Pro stabilizing tail.

What is Semax?

Semax is a lab-made peptide built from a piece of ACTH — a hormone your pituitary gland makes to manage stress. Scientists took the part of ACTH that seemed to influence brain function (without the hormonal cortisol effects) and added a protective tail. The result is a peptide studied for its effects on brain chemistry without triggering the stress hormone cascade.

Mechanism of Action

Semax is hypothesized to enhance BDNF expression, modulate dopaminergic and serotonergic systems, and exert neuroprotective effects through inhibition of oxidative stress. Unlike ACTH, it does not stimulate adrenal cortisol production, allowing researchers to study its neurotrophic effects independently of stress hormone activation.

Research Evidence

Human Evidence

Human Evidence

Semax has been studied in clinical settings in Russia and is approved as an intranasal medication in the Russian Federation for cognitive and neurological applications. Published human studies exist examining its effects in stroke recovery and cognitive performance.

Animal Evidence

Animal Evidence

Extensive animal research has examined neuroprotective effects, learning and memory performance, and brain injury recovery in rodent models.

Laboratory Evidence

In-Vitro / Lab

In vitro research has demonstrated effects on BDNF gene expression, neuronal survival under oxidative stress, and neurotransmitter metabolism.

Frequently Asked Questions

Does Semax raise cortisol like ACTH?

No. Semax is derived from the 4-7 fragment of ACTH specifically because this region does not stimulate adrenal cortisol production. It retains the neurotrophic properties without the hormonal stress effects.

Delivery Methods Studied

  • Intranasal
  • Injectable (subcutaneous)

Stability Notes

Enhanced stability due to the Pro-Gly-Pro C-terminal modification. Intranasal is the primary studied delivery route.
